摘 要:目的:建立高效液相色谱-荧光检测法测定贯叶连翘中金丝桃素的含量。方法:色谱柱为C18柱(5μm,4.6mm×200mm),流动相为甲醇-pH6.5的磷酸盐缓冲液(55∶45),流速为1.0mL.min-1,柱温为30℃,荧光检测波长λEX=580nm,λEM=600nm。结果:该方法的线性范围为2.55~102ng(r=0.9999)。平均加样回收率为101.1%,RSD为1.4%(n=5)。最低检测限为80pg。结论:本方法快速,简单,灵敏度极高。

AbstractObjectiveA HPLC method with fluorescence detection for determination of hypericin in Hypericum perforatum L. was developed.Method:The chromatographic conditions were as follows:a C18 column (5 μm,4.6 mm×200 mm) was eluted with mobile phase consisted of 55% methanol and 45% phosphate buffer (pH=6.5) at the column temperature of 30 .The flow rate was 1.0 mL.min-1 and the fluorescence detector was operated at λex=580 nm and λem=600 nm.Results:The method was proved to be linear in the range of 2.55102 ng with a regression coefficient of 0.999 9.The average recovery was 101.1% with RSD of 1.4% (n=5).The minimum detection limit was 80 pg.Conclusion:The method was proved to be quick,simple and highly sensitive.
